The sample of blood from the patient is lysed and then dissolved into a container that has a smaller container immersed inside it, containing an electrolyte solution. The solution in the smaller container is kept running through two pumps that run out and into the smaller container respectively. One pump runs the solution out of the container and creates a vacuum while the other pump refills the solution into the vacuum creating a continuous flow. The smaller container has an orifice near the container's bottom.
This Hematology Analyzer applies Coulter’s principle by using two electrodes. The internal electrode is contained in the smaller container while the external electrode is within the blood sample solution that is found in the bigger container. The vacuum pulls the sample solution through the orifice (small hole).
In turn, the cell causes resistance (electrical) as it passes via the orifice to the current. The recorded resistance is measured and amplified. The amplified resistance is processed and thereafter processed for interpretation into a histogram by a computer.
